Orally administered oxygen nanobubbles enhance tumor response to sonodynamic therapy

چکیده

Suspensions of oxygen-filled bubbles are under active investigation as potential means relieving tissue hypoxia. Intravenous administration large quantities is, however, undesirable. Previous work by the authors has demonstrated that tumor oxygen levels can be increased following oral phospholipid stabilized nanobubbles. The aim this study was to determine whether would enhance efficacy sonodynamic therapy (SDT), which is known inhibited in hypoxic tissue. Experiments were conducted a murine model pancreatic cancer. Animals treated with SDT (intratumoural injection 1 mM Rose Bengal followed exposure MHz ultrasound, 0.1 kHz pulse repetition frequency, 30% duty cycle, 3.5 W cm−2 for minutes) either or without prior gavage bubbles. A statistically significant reduction rate growth observed groups receiving nanobubbles 5 20 minutes before SDT. Separate measurements using fiber optic probe and expression hypoxia inducible factor (HIF)1α excision, confirmed change levels. These findings offer potentially promising new approach order facilitate cancer therapy.

Orally administered pimonidazole to label hypoxic tumor cells.

Pimonidazole, a "hypoxia marker" normally delivered i.v. or i.p., was instead administered in the drinking water of tumor-bearing mice. As pimonidazole exposure was increased from 3-96 h ad libitum, both the fraction of hypoxic tumor cells and the relative number of pimonidazole adducts in those cells increased. Furthermore, the sustained ingestion of pimonidazole revealed a larger hypoxic frac...

Orally administered Giardia duodenalis extracts enhance an antigen-specific antibody response.

We have identified novel adjuvant activity in specific cytosol fractions from trophozoites of Giardia isolate BRIS/95/HEPU/2041 (J. A. Upcroft, P. A. McDonnell, and P. Upcroft, Parasitol. Today, 14:281-284, 1998). Adjuvant activity was demonstrated in the systemic and mucosal compartments when Giardia extract was coadministered orally with antigen to mice. Enhanced antigen-specific serum antibo...

Antitumor activity of orally administered maitake α-glucan by stimulating antitumor immune response in murine tumor

Maitake α-glucan, YM-2A, isolated from Grifola frondosa, has been characterized as a highly α-1,6-branched α-1,4 glucan. YM-2A has been shown to possess an anti-virus effect in mice; however, it does not directly inhibit growth of the virus in vitro, indicating that the anti-virus effect of YM-2A might be associated with modulation of the host immune system. In this study, we found that oral ad...

Sonodynamic Therapy Using Protoporphyrin IX Conjugated to Gold Nanoparticles: An In Vivo Study on a Colon Tumor Model

Objective(s) Sonodynamic therapy is a physical treatment which utilizes ultrasound waves with an appropriate sensitizer such as protoporphyrin IX (PpIX). The activation of sensitizer depends on cavitation, and therefore, high intensity ultrasound is an important necessity. Beside, high intensity ultrasound can induce side effects on the healthy tissues which have surrounded tumor. The particle...
